My sights aren't quite as high. I'd like to find a truly gem 35-S cent and an equally nice 54-P cent ... ideally in some seller's 3-ring notebook of Lincolns or box of 2-by-2s. Slabbed MS66s that are dull or buckshot or dark or weakly struck or fail to inspire a double-take need not apply.
In over 20 years of looking through raw dealers' stocks, I've found a fair number of gem Lincolns, Jeffersons, and Roosevelts, pre 1965 ... the coins of my youth. The hunt is (almost) everything!

the 1823 and 1827 quarters are certainly up there High end bearded goddess and 1815 half dollars would be on the list A complete 1853 (both arrows and no arrows) mint state set I'm sure there are others, but those would certainly be cool to own, but not worth selling my house for.
The Nova Constellatio Patterns have to be at the top or very near the top. The At*water Wreath Cent, PCGS Sp. 68 Red, and the Mickley Chain Cent are also at the top of my dream list.

In my collection I would really like to get a 1922 no D Lincoln (strong reverse) in AU58 to complete my Dansco Lincoln set. I would not call that my Holy Grail as I could buy one if I was willing to make payments.
For me I will most likely never own, however would be very happy to own, a nice 1876-CC twenty cent piece or a nice 1795 gold Eagle. I do not have a set that requires a Holy Grail to complete so these are high on my want list but will probably never have unless two things happen in proper order. 1) I win a lottery. 2) A nice example comes to market.
Keeping it within reason, a Lafayette Commem Dollar in MS-65. The price has jumped so much in the last 6 years or so they have priced themselves outside my meager income. I shoulda bought one when I could afford it 6 or 7 years ago for around $4,000.00 to $5,000.00.

Tough to say. Right now, I'd say it's the Elizabeth I shilling I misplaced about a year ago and which I have yet to relocate. Certainly I've been trying much harder to find it than anything other coin.
It's not really rare like some of the ones that have been mentioned in this post, but a 1932 D Washington quarter. Everytime I find the one I would like to have I don't have the extra cash for it and there are several available. When I do have the extra cash I can't find even one that suits me.
There are a lot of "Gee, if I won the lottery, then I might actually buy this" coins on my mental list, but if we're talking "Holy Grail", the top-of-the-heap, "what dreams are made of.." kind of coin, it would have to be one I read about and dreamed about as a kid - A Brasher Doubloon! (any variety, I'm not picky!)

However, unless by some miracle I hit the lottery, I don't think I'll be obtaining one anytime soon as they all carry six figure price tags
