Photos aren’t conclusive. Could be a small hair or some fuzz inside the slab. Even if it is a legit strike through it isn’t enough to warrant a premium IMO.

Looks like just a piece of lint which could be from having been stored in one of the sewn cotton 2" x 2" sleeves.
That's a real nice coin! Looks like a strike-through. Does the line appear like a canyon or crevice etched into the coin, or is it raised? That would address the concerns mentioned in the comments above.

If it's incused, which I suspect it is based on your description, then I believe it is a strike-through. Looks like it was from a small thread or fiber ... and I have seen similar examples. Nice looking Quarter as well.

From the pictures I would say struck through lint. I once had a gorgeous proof Trade Dollar that had a large lint strike through in the field that looked like a snake. Probably turned a 66 into a 64.
