Do you think a coin with this black spot on the reverse, belongs in a 66 slab? The coin otherwise is nice in all aspects.

No. However as the second poster wrote, it may have not been there when first graded.
I considered it may have turned while in the slab. I'm surprised by the asking price (2.3X) recommended price, black spot included.
