A gold coin won't have a small spot of toning, and I've never seen milk spots on them before. Since it seems to be strongest at the center, I'd be inclined to say PVC.
People should be required to wear masks when handling unc coins. My guess is a spot of spit from holding the coin raw and having a conversation, and a very small spot of dribble hit the coin, and thats what you get over time.

I believe this is a cosmetic that was used on the surface of the coin to hide problems and this part turned and became visible. It's covering a big hit. Maybe other parts of the coin are affected but don't yet show?
Actually it's a listening device. Since 1933 the federal government is STILL trying to break into coin peoples pockets and so they have installed small listening devices in coins so as to monitor coin peoples activities and create a file on everyone who buys coins for the purposes of confiscating them and taxing them.
